Title of the project.
Inhibition of the HslVU complex of Leishmania as a new therapeutical approach for Leishmaniases : Chemical synthesis and biological evaluation of the inhibitors.

Summary.
Today therapy of Leishmaniases is heavy and complex and there is an urgent need for new targets. The proteasome-like HslVU complex is a promising new target as the inhibition of its expression is lethal for the parasite and is absent in human beings. Two approaches are considered : i) the design of molecules able to inhibit HslVU assembly ; ii) the identification by library screenings and the optimisation of molecules able to inhibit the catalytic activity of HslV protease but not that of proteasome. The post-doctoral fellow will be in charge of the chemical synthesis of molecules as well as their biological evaluation (mainly enzymatic tests), which will be performed in the team of Dr Olivier Coux (CRBM, Montpellier). Another team headed by Prof. Patrick Bastien (Department of Parasitology and Mycology, CHRU, Montpelier) will perform assays on the parasites.

The candidate, possessing a PhD, will be a highly motivated organic chemist. An experience in peptide chemistry and/or solid supported chemistry will be an advantage. An experience in biological testing (enzymatic assays,…) will be another advantage.
