Metallocarbonyl complexes (η5-C5H5)M(CO)n1-N-maleimidato) (M= Fe, W, Mo, Ru ; n= 2, 3) synthesis, reactivity and application in biochemistry

Over the past years we have explored the chemistry of metallocarbonyl complexes of Fe, W, Mo and Ru. The half sandwich complexes display specific and intense absorption bands in IR spectroscopy occurring in the range of 1800 and 2150 cm–1, which is usually free of any absorption of biomolecules or biological matrices. This feature made it useful as a IR-detectable marker to monitor biochemical processes such as hormone–receptor or antigen–antibody interactions. The ethylenic bond of the maleimidato ligand in complexes (η5-C5H5)M(CO)n1-N-maleimidato) (M= Fe, W, Mo, Ru ; n= 2, 3) is readily attacked by nucleophiles such as thiols, amines or imidazole depending on reactional conditions. Recently we have discovered that metallocarbonyl maleimide complexes may also undergo oxa-Michael reaction with compounds bearing hydroxyl groups. All these reactions can be used to introduce metallocarbonyl complexes on various macromolecules e.g. proteins and dendrimers. Metallocarbonyl complexes (η5-C5H5)M(CO)n(η1-N-maleimidato) (M= Fe, W, Mo, Ru ; n= 2, 3) were studied for several biochemical applications including labeling of biomolecules, synthesis of metallocarbonyl functionalized gold and magnetic nanoparticles (to provide IR reporter immunoprobes), enzyme inhibition and CO-releasing molecules (CORMs).
