Design and synthesis of non peptide oligomers for the vectorization of Cathepsin D inhibitors. Potential therapeutic tool in cancerology.

A postdoctoral position funded by a French Research Foundation “Fondation pour la recherche médicale, FRM” will be available for 18 months in the Department of Aminoacids, Peptides and Proteins of the Institute of Biomolecules Max Mousseron (IBMM). The workplace will be the Faculty of Pharmacy of Montpellier within the team « Chemistry of Amino Acids, Peptides, Heterocycles, Supported Chemistry »of IBMM under the supervision of Dr Muriel Amblard.

The main focus of the project is the design of inhibitors targeting Cathepsin D (CathD), a lysosomal enzyme, which is overexpressed and secreted by many solid tumors and implicated in tumor progression and development of metastases as potential anti-cancer agents. The postdoctoral fellow will be involved in the synthesis of bioconjugates, which associate a new family of cell penetrating compounds and potent CathD inhibitors or specific CathD siRNA. The duties will include optimisation of existing lead vectors but also the design of new specific vectors for both CathD inhibitors and CathD siRNA. The conjugates will be tested by the team “Glyco and nanovectors for therapeutic targering” headed by Dr Marcel Garcia at IBMM. 

Qualification of the candidate : Ph.D. in organic chemistry, experience in medicinal chemistry and solid support chemistry will be advantageous but not essential, a history of excellent research productivity. The candidate should also have experience in purification and characterization of organic molecules using analytical techniques like LC-MS, NMR and HPLC. The candidate is expected to be highly motivated and must be independent in designing synthetic routes, solving challenging synthetic problems, efficient in synthesizing targeted molecules, analyzing results to improve drug-like properties of bioconjugates. In addition, the project being at the interface of chemistry and biology, the candidate must be able to closely collaborate with biologists. The candidate should be fluent in French and/or in English.
