Conception, synthèses et propriétés biologiques d’inhibiteurs de l’intégrase du VIH-1

En 2004, nous avons identifié la 2-hydroxyisoquinoléine-1,3-dione comme structure de base à la conception de nouveaux inhibiteurs de l’intégrase du VIH-1. Au cours de ces 8 dernières années, une centaine de composés a été synthétisée. La conception a été basée dans un premier temps en utilisant un modèle moléculaire 3D construit à partir des inhibiteurs disponibles à l’époque, puis en se basant sur les données cristallographiques de l’intasome de l’intégrase du PFV co-cristallisée avec le raltegravir, l’elvitegravir et le dolutegravir.

Une famille de 2-hydroxy-1,3-dioxoisoquinoline-4-carboxamide a été brevetée, un « lead » a été identifié et optimisé en vue d’entrer en phase préclinique.

Ce séminaire permettra de montrer les processus « hit-to-lead » et « lead optimization » au travers d’un exemple et fera l’historique sur l’arrivée sur le marché du raltegravir (Isentress) le 1er inhibiteur de l’intégrase du VIH-1 et de l’elvitegravir (Stribild : Elvitegravir/cobicistat/emtricitabine/tenofovir), partie d’une pilule contenant quatre entités moléculaires différentes.

Design, synthesis and biological properties of HIV-1 integration inhibitors

In 2004, we have identified 2-hydroxyisoquinoline-1,3-dione as a new scaffold for the design of new HIV-1 integrase inhibitors. During these last eight years, about one hundred of new compounds were synthesized. The design was firstly based on a 3D molecular model built from the superimposition of HIV-1 integrase inhibitors, then using the last crystallographic data of the intasome of PFV integrase with raltegravir, elvitagravir and dolutegravir.

A series 2-hydroxy-1,3-dioxoisoquinoline-4-carboxamide was patented, a lead was identified and optimized in order to enter into preclinical phase.

This seminar will show the process from a hit to a lead and the lead optimization through the example of the 2-hydroxy-1,3-dioxoisoquinoline-4-carboxamide series and the history leading to the FDA-approval of raltegravir (Isentress), the first HIV-1 integrase inhibitor and elvitegravir (Stribild : Elvitegravir/cobicistat/emtricitabine/tenofovir) included into a combination pill.
